The C. elegans gene pme-5: molecular cloning and role in the DNA-damage response of a tankyrase orthologue [An article from: DNA Repair]

The C. elegans gene pme-5: molecular cloning and role in the DNA-damage response of a tankyrase orthologue [An article from: DNA Repair] : This digital document is a journal article from DNA Repair, published by Elsevier in 2004. The article is delivered in HTML format and is available in your Amazon.com Media Library immediately after purchase. You can view it with any web browser.

Description:
Tankyrases are recently identified proteins characterized by ankyrin repeats and a poly(ADP-ribose) polymerase (PARP) signature motif. In vertebrates, tankyrases mediate protein-protein interactions via the ankyrin domain. Many partners have been identified that could function in telomere maintenance, signal transduction in vesicular transport, and cell death. To further our knowledge of tankyrases and to study their function in development, we sought and found a tankyrase-related gene in Caenorhabditis elegans that we named pme-5 (poly(ADP-ribose) metabolism enzyme-5). The protein encoded includes a large ankyrin domain and a catalytic PARP domain containing the well-conserved PARP signature sequence and the regulatory region. Unlike other tankyrases, PME-5 lacks a sterile-@a module (SAM), but has a coiled coil domain which may mediate oligomerization. We also found that pme-5 mRNA is alternatively spliced at the fifth exon, producing a long (PME-5L) and a short (PME-5S) transcript. Both isoforms are constitutively expressed during the life cycle of C. elegans. We also show DNA damage increases expression of pme-5, a response that requires the DNA damage checkpoint gene hus-1. Moreover, DNA damage-induced germ cell apoptosis was slightly increased in pme-5(RNAi) hermaphrodites. Altogether, these data indicate that pme-5 is part of a DNA damage response pathway which leads to apoptosis in C. elegans. Late Spring – Criterion Collection

Late Spring – Criterion Collection Serene masterpiece – Westley – Stuck in my head
Director Yasujiro Ozu’s “Late Spring” (1949) tells the story of a widowed Japanese professor and his unmarried 27-year-old daughter, Noriko (Setsuko Hara). Noriko tends to her father and their home, joking that he couldn’t make it without her. The pair seems content with this arrangement, but their friends and relatives begin to note that it’s time for Noriko to marry. Noriko and her father simply laugh off this pressure, but eventually the professor’s sister convinces him that Noriko cannot take care of him forever. Persuading Noriko is more difficult, and the professor may have to deceive her in order to do so.

“Late Spring” depicts the shifting societal mores in post-WWII Japan, as the younger generation started to abandon the traditional Japanese way of life. When Noriko visits her aunt, they sit on the floor wearing kimonos for a formal tea ceremony; when she visits her divorced friend, they sit perched on chairs wearing Western-style clothes and eat shortcake. The contrast is striking. Even the older generation is changing subtly, and Noriko expresses disgust when one of her father’s friends remarries.

The film is a simply-told, serene masterpiece; the deliberate pace allows this unadorned story to unfold realistically. “Late Spring” is not a talky Western-style film filled with self-revelation, confessions, and epiphanies. We never hear directly from Noriko why she seems doesn’t want to marry – we have to discern it from her actions and bits of dialogue. Ozu’s focus on measured character development makes the movie greatly affecting. Some audiences may balk at the slow pace and the formality of “Late Spring.” However, patient audiences will be richly rewarded by this beautiful tale of the sacrifices people make for their loved ones. I think this film is even better than “Tokyo Story,” which many people consider to be Ozu’s finest film.

Image Entertainment: The first of a series of intimate family portraits that would cement Yasujiro Ozu’s reputation as one of the most important directors in cinema history, Late Spring tells the story of a widowed father who feels compelled to marry off his only, beloved daughter. In the hands of two of the director’s finest actors—Chishu Ryu and Setsuko Hara—this poignant tale of love and loss in postwar Japan remains as potent and meaningful today as ever. A masterpiece of postwar Japanese cinema, Yasujiro Ozu’s Late Spring serves as an elegant primer for many of the themes that would define Ozu’s later career. As with other Ozu classics, this is a calm, meditative drama about the dynamics of family, in this case the inevitable separation of 56-year-old father and widower Shukichi (Chishu Ryu) and his adult daughter Noriko (Setsuko Hara), who is content to care for her father and remain unmarried, despite the urging of friends and relatives to find a suitable husband. There are some viable candidates, and several attempts at matchmaking, but the likeliest match is a man who’s already engaged. Noriko simply wishes for things to remain as they are, but when she does eventually marry a handsome chemist who “looks like Gary Cooper,” Ozu’s drama remains intimately focused on the subtle emotions at play; there’s not a scene or sequence that feels out of place, and Late Spring serves a secondary function as a light and lively portrait of post-war Japan, as hints of Western influence (like a Coca-Cola sign in one of the film’s most memorable scenes) that signal Japan’s transition toward a modern commercial economy. Most of all, however, Late Spring is a carefully observed and quietly heartbreaking story of a parent who yearns to set things right for his daughter who must balance her father’s love with her own prospects for a fulfilling future. And while Ozu would go on to examine familial issues in later, equally noteworthy films, Late Spring represents a milestone that would ensure Ozu his rightful place among the greatest of all Japanese directors. –Jeff Shannon

On the DVDs
Criterion’s release of Late Spring contains a few minor flaws in terms of image quality (such as occasional emulsion scratches), but viewers can rest assured that this DVD was mastered from the finest available materials, and the film looks very good considering the conditions of post-war Japan that were typically harsh on films of that period. The “windowbox” framing format accurately preserves the film’s original 1.33:1 aspect ratio. There’s a new and improved English subtitle translation, and the audio commentary by Richard Peña (an Ozu expert and program director of New York’s Film Society of Lincoln Center) emphasizes the literary traditions that inform Ozu’s films, in addition to the director’s signature fixed-camera, low-angle style. Disc 2 includes Tokyo-ga, the 1985 feature by German director (and avid Ozu admirer) Wim Wenders. It’s a tribute to Ozu’s Japan, in which Wenders wanders the city searching for remnants of Tokyo as seen in Ozu’s films, including interviews with Late Spring actor Chishu Ryu and Ozu’s long-time cameraman Yuharu Atsuta. In keeping with Criterion tradition, a 21-page booklet is also included, containing informative essays by critic Michael Atkinson and renowned Japanese-film historian Donald Richie. –Jeff Shannon Late Spring – Criterion Collection

The Bridges of Madison County. A Novel

The Bridges of Madison County. A Novel Magnificent – Der Cubbie –
My first Audio book – but what a great one.

I knew the movie before, but this audio book touched me. Mr. Waller did a great job narrating his book and the love story of a lifetime touched me once again.

I guess, not everybody will likeit, but for me it just great.
What is all the hype about? – –
I read this book because I was tired of seeing it all over the place and assumed that there was something there. I closed it 2 hours later, having finished it in that amount of time, wondering how many poor trees had to die to print this soap opera. Children find out that their mother had an extra-marital affair on their father while they were at a country fair, having left her behind to pick up a wandering photojournalist and they find the romance worthy of celebrating? Yeah! That is reality for you. I am sure there are loads of adult children hoping they found out that their mother was living a lie their entire lives!

Sorry, but I still regret wasting the time on this book. And I won’t see the movie, even on video, because I have already wasted enough time on this sentimental fantasy. I would rather read a Harlequin novel. At least then I would have been forewarned and without excuse for wasting my time.
: Probably the most irresistible love-story formula is that of true love lost, along the lines of Romeo and Juliet, Gone with the Wind, and Titanic. The Bridges of Madison County, a monster hit of the early 1990s, is that kind of torturous melodrama, writ small. Very small. Because lone wolf photographer Robert Kincaid has only four adulterous days to spend with shy Iowa farmhouse wife Francesca Johnson before her husband returns from the state fair with the kids in tow. The novella-sized affair feels much longer than it actually is, mostly because Francesca spends a lot of time gazing at her lover, who is described, by turns, as a “leopard,” “peregrine,” “shaman,” and “star-creature.” The worldly loner melts under this adoration and ends up begging her to run away with him to a safari-adventure life in exotic locales all over the world. “We’ll make love in the desert sand and drink brandy on balconies in Mombasa, watching dhows from Arabia run up their sails in the first wind of morning. I’ll show you lion country and an old French city on the Bay of Bengal.” Alas, Francesca keeps fast to her Midwestern family values. She lets the love of her life (who will, as a result, stay true to her until he draws his last breath) drive away in his truck without her, while leaving it up to her diary to confess the affair to her children after her death. –Sumi Hahn Almquist

National Geographic Investigates: Ancient China: Archaeology Unlocks the Secrets of China’s Past : In 1974, farmers uncover the terra-cotta army of China’s first emperor, Qin Shi Huangdi. Satellite technology reveals about 8,000 soldiers at the tomb complex. Only 1,000 have so far been excavated.

In 1976, archaeologists uncover a Shang Dynasty tomb in the Henan province. They recover artifacts bearing the name Fu Hao. Could this be the same person whose name is found on oracle bones unearthed elsewhere in China?

Bodies encased in suits of jade, and a pristinely preserved mummy are among the other treasures from China’s tombs that continue to provide us with clues to her rich, complex past.

Ancient China includes an interview with Dr. Zhichun Jing, University of British Columbia, and working archaeologist at Anyang, the last capital of the Shang Dynasty. National Geographic Investigates: Ancient China: Archaeology Unlocks the Secrets of China’s Past
